RESUMO

In this study, the effect of environmental aw on microbial inactivation by intense pulsed light (IPL) was investigated. Three different microorganisms (Gram-positive bacteria, Gram-negative bacteria, and yeast) were used as test organisms. The effect of environmental aw was assessed by irradiating each microbial suspension in sodium chloride solutions with different environmental aw levels (0.99-0.80). As the aw decreased, the aggregation of intracellular material of cell interior was changed and the cell number was increased. However, there was no significant difference in microbial reduction according to the aw after the 0.23-3.05 J/cm2 of IPL treatment. It was confirmed that yeast had the highest resistance to IPL because of the differences in cell structure and cell wall components between yeast and bacteria. Additional research is needed to clearly understand the inactivation mechanism according to the type of microorganism by controlling aw using various solutes.

This study evaluated how the efficacy of intense pulsed light (IPL) was influenced by biological factors such as the incubation time and the population of Escherichia coli. According to the 4D value, the microorganisms in the exponential phase were more susceptible to IPL (0.51 J/cm2), while those in the stationary phase were the most resistant (0.67 J/cm2). The microorganisms in the exponential phase could have more critical DNA damage. In addition, the degree of inactivation was affected by the microbial population. When the population was 109 CFU/ml, a maximum 3.4-log reduction was observed after applying IPL at 12.5 J/cm2. In contrast, a population with a density of 1010 CFU/ml showed maximally 0.13-log reduction when IPL was applied at 18.7 J/cm2. This large difference might have been due to cell distribution and aggregation. The study is expected to contribute to the analytical confirmation of the microbial reduction mechanism through non-thermal technologies.

Acrylic acid (AA) chemical that endangers human health through contamination of water, soil, and foods. In this study, the extraction, purification, and detection of AA in various food products were established. The contamination level of AA in food products was investigated as well. Food matrices that were used for method validation were crop, fruit, vegetable, seaweed, beverage, sauce, paste, and pickled food, where the validation was confirmed through cross-checking between two different laboratories by checking the accuracy and precision. Furthermore, sample volume for analysis was optimized. Sonication and syringe filtering were all steps preparing headspace analyzer (HA) before GC-MS analysis. Linearity (R2), limit of detection (LOD), limit of quantitation (LOQ), accuracy and precision of AA, were > 0.99, 0.06-0.17 mg/L, 0.18-0.52 mg/L, 90.97-111.93% and 0.12-9.61 RSD% of intra, inter-day, respectively. White rice sample was the only one sample where AA detected (6.19 mg/L) among 102 food samples. Benzyl chloride is a harmful chemical that contaminates air, water, and food. A static headspace GC-MS method for determining benzyl chloride in food was developed and validated. Two food matrices (fat/oil and chicken) were used for method validation. Sample preparation involved ultrasonication extraction and purification (syringe filtration). Linearity (R 2) was > 0.99, accuracy ranged from 86.91% to 110%, the limit of detection was 0.04-0.17 mg/kg, and the limit of quantification was 0.13-0.52 mg/kg. Recovery varied from 88.19% to 111.77%, and precision ranged from 0.10% to 1.17% in intraday and interday analyses. Among 102 food items (oils, fats, meat, marine, and egg products) distributed in Korea, benzyl chloride was only detected in six of the marine products. The validated analytical method can be used for routine monitoring of benzyl chloride residues in food and thereby prevent the human health risks associated with the consumption of food contaminated with this chemical.

This study quantified six heavy metals (Pb, Cd, As, Sn, Hg, and Me-Hg) in home meal replacement products. Satisfactory linearity (R2 > 0.99), recovery (80.65-118.02%), limits of detection (0.02-2.81 µg/kg), limits of quantification (0.05-8.51 µg/kg), accuracy (80.49-119.87%), precision (0.26-14.93%), standard uncertainty (0.082-0.321%) and relative standard uncertainty (0.084-0.320%) of the six heavy metals were obtained. The average concentration of the six heavy metals was 8.87 µg/kg. Heavy metal concentrations were converted to food intake data of 0.009 µg/kg to recalculate the 95th percentile food intake data (g/day) of individual heavy metals. These were then divided by age group to evaluate the average exposure to heavy metals and determine the 95th percentile of exposure from daily intake and for the whole population, of home meal replacement products. The chronic daily intake amount of six heavy metals was 1.60 × 10-2 µg/kg/day. Based on total chronic daily intake values, the risk and margin of exposure of each of the heavy metals was 9.13 × 107, demonstrating that intake associated with home meal replacement products is negligible.

The aim of this study was to find the optimum condition of pulsed electric field (PEF) and intense pulsed light (IPL) for the enhancement of subcritical water extraction (SWE), which is an eco-friendly extraction method, for extracting tea catechins from green tea leaves (Camellia sinensis). The leaves were treated with PEF under conditions of electric field strength (1, 2 and 3 kV/cm) during 60 s. Moreover, IPL was applied at various voltages (800, 1000, and 1200 V) for 60 s. The SWE was performed for 5 min at varying temperatures (110, 130, 150, 170, and 190 °C). The maximum yield of total catechin was 44.35 ± 2.00 mg/g dry green tea leaves at PEF treatment conditions of 2 kV/cm during 60 s, as well as the SWE temperature of 130 °C. In the case of IPL treatment, the largest amount of total catechin was 48.06 ± 5.03 mg/g dry green tea leaves at 800 V during 60 s when the extraction temperature was 130 °C. The total catechin content was increased by 15.43% for PEF and 25.09% for IPL compared to the value of untreated leaves. This study verified that PEF and IPL had a positive effect on the enhancement of tea catechins extraction from green tea leaves using SWE.

This study investigated how the following four intense pulsed light (IPL) treatment factors affect the inactivation of Bacillus subtilis (KCCM 11,315) spores: distance between the sample and IPL lamp (8, 13, and 18 cm), pulse width (0.5, 1.3, and 2.1 ms), charging voltage (1000, 1200, and 1400 V), and processing time (10, 20, and 30 s). The results showed that all four factors considerably influenced the spore inactivation rate in different ways. Excluding processing time, which does not affect the pulse itself, the effect was largest for pulse width, followed by distance, and charging voltage. The optimal treatment condition that maximized the inactivation rate was a distance of 8 cm, a pulse width of 2.1 ms, a charging voltage of 1000 V, and a processing time of 30 s, which together produced a 6 log reduction. It revealed that individual factors need to be investigated together for achieving the optimal condition of IPL.

Polycyclic aromatic hydrocarbons (PAHs) are carcinogenic and mutagenic compounds that are often formed during the thermal processing of herbal medicine ingredients. In this study, the concentrations of four PAHs (PAH4) in various herbal medicine ingredients were monitored. Further, the QuEChERS method was used to replace conventional pretreatment, a more complex and cumbersome approach. The recovery range of the QuEChERS method ranged between 89.65-118.59%, and the average detection levels of benzo[a]anthracene (BaA), chrysene (CHR), benzo[b]fluoranthene (BbF), and Benzo[a]pyrene (BaP) in 50 herbal medicine ingredients were 0.18, 0.27, 1.13, and 0.17 µg/kg, respectively. The BaP and PAH4 levels in all tested samples were deemed safe according to risk characterization analyses based on European Union and Korean guidelines. Therefore, our findings indicated that the QuEChERS method could be used as an effective alternative to conventional sample pretreatment for the analysis of herbal medicine ingredients.

The purposes of this study were to evaluate the inactivation effects of intense pulsed light (IPL) on indigenous and inoculated microorganisms in fresh and minimally processed foods and the industrial applicability of this nonthermal sterilization method. The samples were treated with IPL by varying the treatment time and voltage. The inactivation effect tended to increase as the treatment conditions increased. Further, indigenous microorganisms showed a lower inactivation level than inoculated microorganisms, E. coli ATCC 25922, due to the variability of indigenous microorganisms and their properties. Chopped garlic showed a higher E. coli inactivation effect (2.65 log reduction after 0.185 J/cm2 of IPL) than peeled garlic (1.21 log reduction) due to its larger surface area. The manila clam showed a lower E. coli inactivation (0.93 log reduction) effect than squid (1.84 log reduction) due to its rougher surface. After the IPL treatment, there was no significant difference in temperature, moisture content, and color.

The objective of this study was to identify whether the efficacy of extracting hesperidin and narirutin from Citrus unshiu peel by-products can be increased by combining pulsed electric field (PEF) and subcritical water extraction (SWE). The samples were treated with a PEF at a strength of 3 kV/cm for 60 and 120 s. Subsequent SWE was conducted at extraction temperatures of 110-190 °C for 3-15 min. The concentration of hesperidin was highest at 46.96 ± 3.37 mg/g peel (dry basis) after PEF treatment at 120 s, combined with SWE at 150 °C for 15 min, while that of narirutin peaked at 8.76 ± 0.83 mg/g after PEF treatment at 120 s, integrated with SWE at 190 °C for 5 min. The concentrations of both hesperidin and narirutin increased with PEF treatment time. The PEF increased the amounts of hesperidin and narirutin extracted by 22.1% and 33.6%, respectively. This study demonstrate the potential of PEF pretreatment for enhancing the SWE of flavonoids from C. unshiu peel.

It is important to inactivate spore-forming bacteria in foods because their spores are highly resistant to various stresses. Although thermal treatment is an effective inactivation method, the associated high temperatures can cause changes in food quality. Intense pulsed light (IPL) is a nonthermal technique that can effectively improve food safety. This study evaluated the inactivation effects of IPL at various fluences on Bacillus subtilis spores. IPL treatment at a total fluence of 7.40 J/cm2 resulted in a 7 log reduction, indicating the potential of IPL to effectively inactivate bacterial spores. The sensitivity of B. subtilis spores to IPL during germination and outgrowth was also measured. The resistance to the IPL increased temporarily until 1 h after the start of incubation, and then gradually decreased for longer incubation periods. This temporary increase in resistance at the early stage of incubation was attributed to the leakage of dipicolinic acid from the spores. The results also showed that the inactivation efficiency increases after 1 h pre-incubation because the numbers of vegetative cells increased with the incubation time.

This study investigated the microbial inactivation effects of intense pulsed light (IPL) treatment as an alternative to chemical treatment for decontaminating the radish and pak choi seeds. The f R values (which indicate the resistance to IPL treatment) for radish and pak choi seeds were 24.50, 20.81 J/cm2, respectively. This resistance exhibited by seeds to IPL treatment is related to their surface roughness. Their Rq (the root-mean-square roughness), average surface roughness (Ra), and 10-point height roughness (Rz) values indicate that each crevice on a rough surface could shelter microorganisms from IPL. Viability tests of seeds exposed to IPL treatment indicated that the average germination rates of treated seeds exceeded 85% on day 3 of germination, which is considered as an acceptable criterion for germination. Also, on day 5 of germination the average shoot lengths of sprouts exposed to IPL did not differ significantly from those of untreated seeds.

We conducted this study to identify the factors for predicting poor outcomes in chronic myeloid leukemia patients who failed to achieve a 3-month early molecular response (EMR). Of the 413 newly diagnosed, chronic phase, chronic myeloid leukemia patients receiving imatinib (IM), 120 (29.1%) failed to achieve a 3-month EMR. With a median follow-up of 67.0 months, 39 patients continued IM treatment with at least complete cytogenetic response (CCyR), and 81 patients permanently discontinued IM treatment. The cumulative incidence rates of CCyR and major molecular response (MMR) by 3 years were 90.1 ± 3.9% and 53.7 ± 7.3%, respectively. After adjusting for potential factors, multivariate analyses showed that a transcript type of e13a2, compared with e14a2, and a larger spleen size were independent factors for failure of overall MMR. The predictive factors outlined in this study may provide valuable information for high-risk patients who would benefit from early decision-making regarding therapy change.

BACKGROUND/AIMS: The aim of this study was to identify the role of BCR-ABL1 transcript level as a predictor for post-transplant relapse and outcome in patients who underwent allogeneic stem cell transplantation (SCT) for chronic phase (CP) chronic myeloid leukemia (CML). METHODS: Of 101 patients receiving allograft in CML CP, 85 had available quantitative reverse transcriptase polymerase chain reaction data at post-transplant 3 months. These patients were divided into two groups according to molecular response (MR4.5), defined as a BCR-ABL1 transcript level ≤ 0.0032% on the international scale, at 3 months based on receiver operating characteristic curve analysis of relapse. RESULTS: The 4-year overall survival and event-free survival (EFS) were 80.6% and 57.3%, respectively, and the cumulative incidence of relapse at 4 years was 29.6% after a median follow-up of 126.4 months. We performed multivariate analyses including potential variables to evaluate the early predictive role of MR4.5 at 3 months and found that MR4.5 at 3 months was associated with a higher EFS (p = 0.028) and showed a trend for a lower relapse rate (p = 0.089). CONCLUSIONS: our results imply that frequent molecular monitoring and immune suppressive therapy modulation are required for patients without reduction of BCR-ABL1 transcripts to this level after SCT.

The sporicidal activities against Bacillus subtilis spores of surfactant components with hydrophilic and hydrophobic properties that can lead to the denaturation of various proteins comprising the spore structure were investigated. The reduction in spore numbers by each of the surfactant components bornyl acetate, geranyl acetate, pinene, p-cymene, camphene, citral, 2,3-dihydrobenzofuran, polylysine, and thiamine dilaurylsulfate at 1% was estimated at 1 to 2 log CFU/ml. The average hydrophilelipophile balance value of surfactants with sporicidal activity causing a reduction of 1 to 2 log CFU/ml was 9.3, with a range from 6.7 to 15.8, which is similar to the values of various chemical surfactants of 9.6 to 16.7. The results also showed that the surfactants that were hydrophobic were more effective than those that were hydrophilic in killing B. subtilis spores. Furthermore, the sporicidal effect of surfactants like geranyl acetate and γ-terpinene was significantly enhanced in the presence of a germinant, because L-alanine and synergistic cofactors (e.g., K(+) ions) trigger cortex hydrolysis in spores.

To explore the factors for achieving early molecular responses (EMR; BCR-ABL1 ≤10% at 3 months, ≤1% at 6 months) by imatinib (IM), baseline characteristics including individual BCR-ABL1 transcript level, dose intensity, and IM trough level on day 29 were analyzed in 286 chronic phase chronic myeloid leukemia patients. Distinct predictive factors for achieving EMR at 3 months and 6 months were noted. Blast count at diagnosis and IM trough level on day 29 were significantly associated with an achievement of 3-month EMR. Early decline of BCR-ABL1 transcript, low Sokal risk, and mean daily dose (≥350mg/day) by 6 months were associated with an achievement of 6-month EMR. Understanding the predictive factors for EMR may provide additional information to guide clinical decisions on the changing therapies at each landmark.

This research aims to develop a questionnaire of deviant behavior for the Korean elderly people which may make a big contribution to the examination of deviance behavior of the elderly people and may play an important role in providing a methodological basis. In order to accomplish the purpose of the this study, there were three different stages; (a) making preliminary question items, (b) refining the items of the scale through a plot study, and (c) finalizing question items by a main survey. In the first stage, 43 question items were developed using the open-ended questionnaire and structural inquiry of succession from 137 elderly people who are over 65 yr. In the second phase, based on data collected by the 200 elderly people pilot testing was performed through exploratory factor analysis and reliability test. The scale is a 27-item self-report questionnaire. In the main survey conducted by 184 elderly people, 21 items, which consisted of four subfactors, were finalized in order to measure deviance behaviors of the Korean elderly people: social deviance (n=8), economic deviance (n=5), psychological deviance (n=5), and physical deviance (n=3).
